dc.description.abstract | Die Arbeit befasst sich mit der oxidativen Phenolkupplung des Siderins (4,7-Dimethoxy-5-methylcumarin) und seiner Derivate in Aspergillen. Der Schwerpunkt liegt dabei in der Aufklärung der Biosynthese von Kotanin, dem 8,8'-Dimer des Siderins, das von Aspergillus niger regio- und stereoselektiv synthetisiert wird. Es wurde ein chemischer Zugang sowohl zu den monomeren als auch den dimeren Cumarinen etabliert. Hierauf aufbauend wurde ein analytischer HPLC-Assay entwickelt, der die Analyse von Rohextrakten verschiedener Pilzstämme erlaubt. Durch Verimpfung von 13C-markierten Monomeren konnte die oxidative Phenolkupplung in A. niger eindeutig bewiesen werden. Darüber hinaus gaben Verfütterungsexperimente mit Verbindungen, die sowohl an der Carbonylgruppe als auch an Methoxygruppen 13C-markiert waren, einen Einblick in die Reihenfolge von O-Methylierungs- und Phenolkupplungsschritten bei der Kotaninbiosynthese. Für die Identifizierung der entsprechenden enzymatischen Aktivität wurde eine Cosmidbank von A. niger angelegt, die in geeignete Aspergillus-Wirtsstämme transformiert und so auf Metabolitproduktion gescreent werden kann. Chemische Synthese Ausgehend von 4-O-Methylorsellinsäuremethylester und seinen regioisomeren Dimeren (3,3';5,5';3,5'), die leicht über eine oxidative Phenolkupplung des Orsellinsäureesters erhältlich sind, wurden die die Cumarine Siderin, Kotanin, Isokotanin A (6,6'-Bisiderin) und Desertorin C (6,8'-Bisiderin) in einer neuen und hocheffizienten Dreistufenstrategie dargestellt. Zusätzlich wurden beide Atropisomere des Kotanins aus den hochreinen Atropisomeren des 3,3'-dimeren Orsellinsäureesters erhalten. Über diesen synthetischen Ansatz sind außerdem alle Hydroxyderivate von Siderin leicht zugänglich. 13C-Markierungen sowohl an den Methoxygruppen als auch am Kohlenstoffskelett (Carbonylisches C-Atom) können leicht und effektiv eingebaut werden. Analytik Unter Verwendung der chemisch hergestellten Verbindungen wurde ein optimierter HPLC-Assay etabliert, der eine schnelle und einfache Untersuchung von Rohextrakten verschiedener Pilze auf Cumarine ermöglicht. Darüber hinaus wurde eine Methode entwickelt, die das Screenen einer Pilz-Genbank (mehrere tausend Stämme) erlaubt. Die Cumarine wurden aus den Pilzen isoliert und charakterisiert (NMR, MS, IR, UV). Die absolute Konfiguration der Bicumarine wurde durch CD-Spektroskopie verifiziert. Biosyntheseuntersuchungen Aus 13C-Markierungsexperimenten kann folgender Biosyntheseweg für Kotanin hergeleitet werden. Das Polyketid 4,6-Dihydroxy-5-methylcumarin wird zu Desmethylsiderin (4-Methoxy-7-hydroxy-5-methylcumarin) O-methyliert. Es folgt ein stereoselektiver oxidativer Phenolkupplungsschritt zum Orlandin (8,8' dimeres Desmethylsiderin), das über Desmethylkotanin zu Kotanin methyliert wird. Molekularbiologische Arbeiten Zur Identifizierung des enzymatischen Systems, das die oxidative Phenolkupplung katalysiert, wurde zunächst eine Cosmidbank von Aspergillus niger erstellt. Der "Cosmid-Shuttle-Vektor" enthält ein Hygromycin-Resistenz-Gen, das eine Transformation in verschiedene Aspergillen und anschließendes Testen auf biosynthetische Aktivität erlaubt. Da die Cumarine polyketidischen Ursprungs sind und PKS-Gene in filamentösen Pilzen gewöhnlich geclustert vorliegen, wurde die Cosmidband auf "nicht-reduzierende" PKS-Gene gescreent, um so gegebenenfalls einen Polyketid-Biosynthesecluster zu identifizieren, der die Aktivität zur Phenolkupplung kodiert. | |
dc.description.abstract | Research on the stereoselective oxidative phenolic coupling of coumarins in Aspergillus niger Biarylic natural products have been isolated from diverse organisms such as bacteria, fungi, plants and insects. An oxidative phenol coupling step is generally assumed for their biosynthesis. However, the knowledge on the corresponding enzymatic activities is still very poor. From the chemical point of view the catalytic regio- and stereoselective phenol coupling is very interesting too, since biaryl syntheses usually require demanding multistep procedures if stereoselectivity is required. This research project deals with the oxidative phenol coupling of siderin (4,7-dimethoxy-5-methylcoumarin) and its derivatives in Aspergillus spp. We focused on Aspergillus niger producing regio- and stereoselectively kotanin, the 8,8' dimer of siderin. First, an efficient chemical access to both the monomeric and dimeric coumarins was established. With these compounds in hand an optimized HPLC assay has been set up, which also allows a screening of many different fungal strains. The existence of oxidative phenol coupling in A. niger was confirmed by feeding experiments with 13C-labelled monomers. Furthermore, the application of compounds labelled both at the carbonyl C-atom and the methoxy-group gave an insight into the order of O-methylation and oxidative coupling steps in kotanin biosynthesis. For the identification of the corresponding enzymatic activity a cosmid library of Aspergillus niger has been generated, which allows a transformation into Aspergillus host strains and further screening for metabolite production. Chemical synthesis Starting from methyl 4-O-methylorsellinate, and its regioisomeric dimers, easily available by an oxidative coupling reaction of methyl orsellinate, the natural occurring coumarins siderin, kotanin, isokotanin A (6,6'-bisiderin) and desertorin C (6,8'-bisiderin) have been synthesised in a new and highly efficient three step transformation. In the case of kotanin both atropisomers have been prepared from the pure atropisomers the 3,3' dimeric orsellinate. Additionally all kind of hydroxy derivatives of the monomer siderin are available using this synthetic approach and 13C-labels can be introduced in a straightforward manner at the carbon skeleton (carbonylic C-atom) as well as at the methoxy groups. Analytics Using the chemically synthesised compounds an optimised HPLC-assay has been set up, which allows screening of the raw extracts of many fungal strains. Additionally a modified HPLC method has been established for a screening of fungal libraries in an adequate period. The natural products were isolated from the fungus and were characterised (NMR, MS, IR, UV). The absolute configuration of the bicoumarins was determined by CD spectroscopy. Biosynthetic research The biosynthetic pathway for kotanin can be derived from 13C-labeling experiments as follows: The polyketide 4,7-dihydroxy-5-methylcoumarin is O-methylated to demethylsiderin (4-methoxy-7-hydroxy-5-methylcumarin), followed by a regio- and stereoselective oxidative phenol coupling step to orlandin (8,8' dimeric demethylsiderin). This is O-methylated to desmethylkotanin and then to kotanin. Molecular biology For the identification of the enzyme(s) catalysing the oxidative phenol coupling reaction a cosmid library of Aspergillus niger has been generated. The cosmid shuttle vector contains a hygromycin resistance gene, which allows a transformation into different Aspergillus strains and testing for biosynthetic activity. Since the coumarins have a polyketidic origin and PKS-genes are usually clustered in filamentous fungi, the cosmid library was screened for 'non reducing' PKS-genes in order to identify a cluster which encodes the phenol coupling activity. | |
