dc.description.abstract | Method development for enantioselective separation of polyphenols in different matrices New methods for the chiral separation of polyphenols in different matrices by capillary electrophoresis (CE) as well as by high performance liquid chromatography (HPLC) were developed. The methods are not limited to foodstuffs; they also cover biological matrices. Human plasma after consumption of cocoa was analysed to detect (+)/(–)-catechin and (+)/(–)-epicatechin. For the chiral analysis of the flavan-3-ols in human plasma, a HPLC-CEAD-System (HPLC with coulometric electrode array detector) was used. The CEAD is highly sensitive and is able to detect polyphenols in concentrations of a few ng/mL. Using a permethylated-γ-cyclodextrin HPLC-Column, a chiral HPLC method was developed for the flavan-3-ol enantiomeres catechin and epicatechin. After a suitable sample preparation method for human plasma with enzymatic hydrolysis of the glucuronidated and sulfated metabolites, it was possible to separate the monomeric flavan-3-ols. This method shows high recovery, good reproducibility and low limits of detection (LOD) and limits of quantification (LOQ). In a self-experiment, it could be noticed that both enantiomeres of catechin were absorbed from human organism. Two further methods were developed: 1. a new method for chiral separation of flavanonglycosides in citrus juice by CE with “sample stacking”. Thereby, it was possible to chirally separate four flavanonglycosides (hesperidin, didymin, narirutin and narirutin-4´-glycoside) in orange juice. 2. a cyclodextrin modified micellar electrokinetic chromatography method (CD-MEKC) to chirally separate flavan-3-ols in cocoa and cocoa products. The CD-MEKC was compared to an already existing CZE Method [Kofink M. 2009] for chiral separation of (+)/(–)-catechin and (+)/(–)-epicatechin. Both methods provide the same results but the CD-MEKC method shows better resolution. | |
