dc.description.abstract | iNOS-abhängige NO-Synthese in Makrophagen ist entscheidend von der zellulären L-Arginin-Aufnahme abhängig. Es zeigt sich in diesen Zellen eine parallele Regulation von iNOS und L-Arginin-Aufnahme. So führen Stimuli, die eine Induktion der iNOS-Expression bewirken stets zu einer Hochregulation des L-Arginin-Transportes und verstärken die Expression des kationischen Aminosäuretransportes CAT-2B. Andererseits reduzieren Mediatoren und Medikamente, welche eine Inhibition der iNOS-Expression bewirken, ebenfalls die CAT-2B-Expression. Für NF-kB ist eine Rolle als essentieller, allein jedoch nicht suffizienter, Transkriptionsfaktor für die Expression von sowohl iNOS als auch des CAT-2-Gens belegt. Im Maus-iNOS-Promotor wurde ein LPS-responsives Element (LREAA) in der Nähe der NF-kB-Bindungsdomäne als zusätzliche essentielle regulatorische Domäne identifiziert (Xie 1997, J. Biol. Chem. 272:14867). Es wurde eine mögliche Rolle von LREAA für die Regulation der Expression von iNOS und CAT-2B in Rattenalveolarmakrophagen mit Hilfe eines Oligodesoxynukleotid-Decoy-Ansatzes untersucht. Die Expression von mRNA für iNOS und CAT-2B in Rattenalveolarmakrophagen ist durch AP-1 und LREAA-decoy-ODN verschieden beeinflußt. Dies ist ein Hinweis, dass die Regulation der Expression von iNOS und CAT-2B von verschiedenen zellulären Signalen kontrolliert wird, trotz der Tatsache, das die Expression von iNOS und CAT-2B in Rattenalveolarmakrophagen durch verschiedene Mediatoren und Medikamente (wie z. B. LPS, Interferon-gamma, Polyamine und Glucocorticoide) in paralleler Weise beeinflußt wird. | en |
dc.description.abstract | Differential role of LPS-responsive element AA (LREAA) for the regulation of the expression of iNOS and CAT-2B in rat alveolar macrophages iNOS-mediated NO synthesis in macrophages depends critically on the cellular uptake of L-arginine. There appears to be a parallel regulation of iNOS and the L-arginine transport in these cells. Thus, stimuli inducing iNOS expression allways cause an upregulation of L-arginine transport and enhance the expression of the cationic amino acid transporter CAT-2B. On the other hand, mediators and drugs causing inhibition of iNOS expression also reduce CAT-2B expression. NF-kB has been shown to be an essential, but alone not sufficient transcription factor for the expression of both the iNOS and CAT-2 gene. In the mouse iNOS promoter in vicinity of NF-kB-binding domain, a LPS responsive element (LREAA) has been identified as an additional essential regulatory domain (Xie 1997, J. Biol. Chem. 272:14867). A possible role of LREAA for the regulation of the expression of iNOS and CAT-2B in rat alveolar macrophages was tested using an oligo-desoxy-nucleotide (ODN) decoy approach. The expression of mRNA for iNOS and CAT-2B in rat alveolar macrophages is differentially affected by AP-1 and LREAA decoy ODN indicating that the regulation of iNOS and CAT-2B expression is controlled by different cellular signals, despite the fact that the expression of iNOS and CAT-2B in rat alveolar macrophages is affected by various mediators and drugs (such as LPS, interferon-gamma, polyamines and glucocorticoides) in a parallel manner. | en |
