dc.description.abstract | Secreted class-III semaphorins exert their effects in axon guidance and neuronal migration by binding with receptors, such as plexins and neuropilins. Neuropilins are insufficient to convey signals of their own; rather, they form complexes with plexins to propagate signals of semaphorins into the cells. Though the role of class-III semaphorins in governing fasciculation, axon growth and cell migration has been studied previously, it is far away from our understanding how their receptors (plexin-As and neuropilins) take part in the axonal guidance of cranial and spinal motor neurons. It has been demonstrated that plexin-A2 and neuropilin-2 (Npn-2) control the motor somal positioning in the chick spinal cord. However, it is still unknown whether they are involved in the regulation of cranial motor neurons. For this purpose, we first analyzed the expression of plexin-A1, plexin-A2, plexin-A4 and Npn-1 and Npn-2 in the motor neuronal groups within the chick hindbrain. Our results demonstrated that all analyzed plexins and neuropilins were selectively expressed by hindbrain motor neurons. For instance, plexin-A1, plexin-A2 and Npn-1 were expressed by both dorsal and ventral exiting cranial motor neurons, whereas plexin-A4 and Npn-2 only by dorsal exiting cranial motor neurons. Based on the expression data, we selected plexin-A2 and Npn-2 genes for knockdown experiments by in ovo-electroporation of short hairpin RNA (shRNA) constructs into the ventral neural tube at the post-otic hindbrain level, from which motor neurons of the vagus (nX), accessory (nXI) and hypoglossal (nXII) nerves originated. Unlike the spinal cord, where loss of function of either plexin-A2 or Npn-2 induced ectopic migration of motor neuron somata along the ventral root, only Npn-2 in the hindbrain induced ectopic migration of motor somata but along the dorsal root. In addition, inhibition of function of Npn-2 resulted in misrouting and severe defasciculation of dorsal exiting (vagus and accessory) motor axons. Furthermore, knockdown of plexin-A2 led to the significant (P<0.001) reduction of motor neuron population in the ventral neural tube and impaired fasciculation of ventral exiting (hypoglossal) motor axons. These results indicate that plexin-A2 and Npn-2 act independently in the axonal guidance of cranial nerves in chick embryos. | |
dc.description.abstract | Die Rolle von Plexin-A2 und Neuropilin-2 bei der axonalen Wegfindung von Hirnnerven im Vogelembryo Sezernierte Klasse-III Semaphorine entfalten ihre Wirkung auf die Wegfindung der Axone und die Wanderung der Neurone durch Bindung an Rezeptoren wie z.B. die Plexine und Neuropiline. Damit Semaphorine ein Signal in die Zelle hinein abgeben können, reicht die Bindung an Neuropiline nicht aus, vielmehr müssen diese dafür einen Komplex mit Plexinen bilden. Obwohl die Rolle der Klasse-III Semaphorine bei der Bündelung und dem Wachstum der Axone wie auch der Zellmigration bereits vielfach untersucht worden ist, ist noch wenig über Bedeutung der Rezeptoren (Plexine-A und Neuropiline) für die Wegfindung der Axone kranialer und spinaler Motoneurone bekannt. Es ist bereits gezeigt worden, dass Plexin-A2 und Neuropilin-2 (Npn-2) die Lage der motorischen Somata im Rückenmark des Hühnchens kontrollieren, es war aber nicht bekannt, ob dies auch für die kranialen Motoneurone gilt. Daher analysierten wir zunächst die Expression von Plexin-A1, Plexin-A2, Plexin-A4 sowie Npn-1 und Npn-2 in Motoneuron Gruppen des Rhombencephalons des Hühnchens. Unsere Ergebnisse zeigten, dass alle Plexine und Neuropiline selektiv in bestimmten Motoneuronen exprimiert werden. So werden zum Beispiel Plexin-A1, Plexin-A2 und Npn-1 von Motoneuronen exprimiert, die sowohl dorsal wie auch ventral austreten, während Plexin-A4 und Npn-2 nur von dorsal austretenden Motoneuronen exprimiert werden. Auf der Grundlage dieser Daten haben wir zwei Gene ausgewählt, Plexin-A2 und Npn-2, mit denen knockdown Experimente durch in ovo- Elektroporation von short hairpin RNA (shRNA) in das ventrale Neuralrohr in Höhe des post-otischen Rhombencephalons durchgeführt wurden, wo die Motoneurone des Nervus vagus, des Nervus akzessorius und des Nervus hypoglossus entstehen. Anders als im Rückenmark, wo die Ausschaltung von Plexin-A2 wie auch Npn-2 ektopische Migration der neuronalen Somata entlang der ventralen Wurzeln induziert, verursachte lediglich der Verlust von Npn-2 eine ektopische Migration, allerdings entlang der dorsalen Wurzel. Darüber hinaus verursachte die Hemmung von Npn-2 eine Fehlleitung und deutliche Aufhebung der Bündelung dorsal austretender Axone des Nervus vagus und Nervus akzessorius. Demgegenüber führte ein knockdown von Plexin-A2 zu einer signifikanten Reduktion der Anzahl von Motoneuronen im ventralen Neuralrohr (P<0.001) und beeinträchtigte die Bündelung ventral austretender motorischer Axone des Nervus hypoglossus. Diese Befunde zeigen, dass Plexin-A2 und Npn-2 unabhängig voneinander die axonale Wegfindung kranialer Nerven im Hühnchen Embryo beeinflussen. | |
